P26 Creating database for identification of marine bacteria using MALDI-TOF MS

PCR based 16S rRNA gene analysis for taxonomic identification of bacteria is time-consuming and costly and today matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) is established as a rapid and cost effective alternative in clinical laboratories. However, the current databases in the market offer limited diversity of environmental bacteria.

The aim of this ongoing study is to create a database for identification of bacteria isolated from marine invertebrates using MALDI-TOF MS. High quality main spectra (MSP), using a fast protein extraction method and the MALDI Biotyper CA System, are generated for constructing a new reference database. These MSPs are based on multiple measurements of single, defined strains, to ensure that the true biological variability of an organism is captured. As new MSPs of bacteria are uploaded, this database will be a valuable tool for rapid identification of unknown cultivable marine bacteria.
